目的 :探讨血府逐瘀汤对用血瘀证兔模型血清损伤内皮细胞的保护作用 ,阐明该方调节血管内皮细胞内分泌功能的作用机理。方法 :将正常组、血瘀模型组、血瘀 +中药组和高脂血症组兔血清加入同批培养的正常兔主动脉内皮细胞中 ,作用2 4h后 ,观察细胞液中ET、NO的含量和t PA、PAI及AT Ⅲ活性的变化。结果 :与正常血清组相比 ,血瘀血清组细胞液中的ET和NO含量都明显升高 ,PAI活性也明显升高 ,AT Ⅲ活性则明显降低 ,t PA活性虽有下降趋势 ,但无显著性意义 ;而血瘀 +中药血清组细胞液中的ET和NO含量以及t PA、PAI和AT Ⅲ活性都未有明显改变。结论 :血府逐瘀汤能调节血瘀证兔模型血清对内皮细胞ET/NO的释放平衡作用和降低其对抗凝、纤溶功能的影响 ,对内皮细胞起到一定的保护作用。
Objective: To investigate the protective effect of Xuefu Zhuyu Decoction on endothelial cells injured by blood stasis model in rabbits, and to elucidate the mechanism of this prescription in regulating the endocrine function of vascular endothelial cells. METHODS: Normal, blood stasis model group, blood stasis + Chinese medicine group and hyperlipidemia group rabbit serum were added into normal rabbit aortic endothelial cells cultured in the same batch. After 24 hours, the ET and NO in the cytosol were observed. Content and changes in t PA, PAI, and AT III activity. RESULTS: Compared with the normal serum group, the contents of ET and NO in the serum of blood stasis serum group were significantly increased, the activity of PAI was also significantly increased, the activity of AT III was significantly decreased, and the activity of t PA was decreased, but there was no Significant significance; and blood stasis + Chinese medicine serum ET and NO levels in the cell fluid and t PA, PAI and AT III activity did not change significantly. Conclusion : Xuefu Zhuyu Decoction can regulate the balance of ET/NO release from endothelial cells and decrease the anticoagulant and fibrinolytic function in serum of rabbits with blood stasis syndrome, and it can protect endothelial cells.
