目的:比较苯酚-硫酸法与硫酸-紫外法在测定千两茶多糖含量上的差异。方法:千两茶粉经水提醇沉,Sevage法除蛋白后,采用苯酚-硫酸法和硫酸-紫外法测定茶多糖含量。结果:苯酚-硫酸法标准曲线方程为Y=4.755 0X-0.029 5(R2=0.990 4),硫酸-紫外法标准曲线方程为Y=4.770 0X-0.065 1(R2=0.993 1),均在0.02~0.1 g·L-1有良好的线性关系。所测茶多糖质量分数分别为2.00%,3.21%,其差异有统计学意义。结论:硫酸-紫外法作为一种多糖检测新手段,可简便测定多糖含量。
OBJECTIVE: To compare the differences between the phenol-sulfuric acid method and the sulfuric acid-ultraviolet method in the determination of the content of polysaccharides in two-stem tea. Results The standard curve equation of phenol-sulfuric acid method was Y = 4.755 0X-0.029 5 (R2 = 0.990 4). The measured tea polysaccharides mass fraction were 2.00%, 3.21%, the difference was statistically significant. Conclusion: Sulfuric acid - ultraviolet method as a new means of polysaccharide detection, can be easily determined polysaccharide content.
