目的:研究冷冻解冻对大肠埃希菌超微结构的损伤和修复。方法:将正常的和经冷冻解冻损伤和修复后的大肠埃希菌作负染色和超薄切片,在电子显微镜下观察其超微结构的变化。结果:(1)冷冻解冻破坏大肠埃希菌的超微结构,细胞壁、细胞膜损伤,胞内有空泡形成;(2)修复后的大肠埃希菌,其超微结构基本恢复正常;(3)冰晶的形成、胞质浓缩产生的盐害是冷冻损伤的主要机理。结论:冷冻解冻可破坏大肠埃希菌的超微结构。
Objective: To study the damage and repair of Escherichia coli ultrastructure by freezing and thawing. Methods: The normal and frozen thawed and repaired Escherichia coli negative staining and ultrathin sections, the ultrastructure of the changes observed under an electron microscope. Results: (1) Frozen thawing destroys the ultrastructure of Escherichia coli, cell wall, cell membrane damage, intracellular vacuolar formation; (2) the repair of Escherichia coli, its ultrastructure basically returned to normal; (3) ) The formation of ice crystals, the salt damage caused by cytoplasmic condensation is the main mechanism of freeze injury. Conclusion: Frozen thawing can destroy the ultrastructure of Escherichia coli.
