目的体外观察选择性环氧合酶-2(COX-2)抑制剂NS-398对胰腺癌细胞株PC-3增殖的影响,并对其可能机制进行探讨。方法体外培养胰腺癌细胞株PC-3,予以不同浓度NS-398处理,以MTT试验检测细胞生长情况;以流式细胞术分析细胞凋亡情况以及细胞周期再分布情况。结果MTT试验结果显示NS-398可抑制胰腺癌细胞增殖:流式细胞术分析显示NS-398处理组细胞凋亡率明显增高,并形成G1期、M期细胞数目明显增多,S期细胞数目减少的细胞周期再分布。结论选择性COX-2抑制剂NS-398可以抑制胰腺癌细胞株PC-3细胞的增殖,其机制与改变细胞周期分布、促进细胞凋亡有关。
Objective To investigate the effect of selective cyclooxygenase-2 (COX-2) inhibitor NS-398 on the proliferation of pancreatic cancer cell line PC-3, and to explore its possible mechanism. Methods Pancreatic cancer cell line PC-3 was cultured in vitro and treated with different concentrations of NS-398. Cell growth was detected by MTT assay. Apoptosis and cell cycle redistribution were analyzed by flow cytometry. Results The results of MTT assay showed that NS-398 inhibited the proliferation of pancreatic cancer cells. Flow cytometry analysis showed that the apoptosis rate of NS-398 treated group was significantly increased, and the number of cells in G1 and M phases increased significantly, and the number of cells in S phase decreased. The cell cycle is redistributed. Conclusion Selective COX-2 inhibitor NS-398 can inhibit the proliferation of pancreatic cancer cell line PC-3. The mechanism is related to the change of cell cycle distribution and apoptosis.
