目的应用DNA条形码技术对鼠疫宿主动物进行分子生物学的鉴定。方法运用PCR方法检测锡林郭勒盟9种啮齿动物的149条线粒体COI基因657 bp的片段序列,用MEGA 5.0软件采用邻接法(NJ)构建分析系统发育树。结果 NJ进化树能清楚地区分9个不同的类群。结论通过COI基因应用DNA条码技术可以对锡林郭勒盟的鼠疫宿主动物进行鉴别,同时可为形态鉴别提供佐证数据。
Objective To identify the molecular biology of plague host animal using DNA barcode technology. Methods A total of 149 mitochondrial COI gene fragments of 657 bp in nine rodents of Xilin Gol League were detected by PCR. The phylogenetic tree was constructed by using neighbor-joining (NJ) software with MEGA 5.0 software. Results The NJ phylogenetic tree clearly distinguished nine different taxa. Conclusion The application of DNA barcode technology in COI gene can identify plague host animals in Xilin Gol League and provide supporting data for morphological identification.
