目的 获得旋毛虫成虫期特异性基因序列。方法 应用旋毛虫成虫期特异性探针对成虫cDNA文库进行杂交筛选、克隆测序及核苷酸序列数据库 (GenBank)查寻比较 ,采用DNAstar软件进行基因序列分析。结果 利用消减杂交 (subtractedhybridization)获得的旋毛虫成虫期特异性探针筛选成虫cDNA文库 ,获得一个长 16 2 9bp的基因。其开放阅读框架由 146 4个核苷酸组成 ,编码 487个氨基酸。序列分析表明 ,该序列是一个尚未见报道的旋毛虫基因序列 ,在 2 0 7~ 2 70位氨基酸间存在两个锌指结构 ,在 5 2~ 6 4,10 8~ 116 ,137~ 16 3,2 2 6~ 2 6 0位氨基酸处存在预测的抗原表位。结论 获得旋毛虫成虫期特异性基因 ,其编码的蛋白具有一定的抗原表位
Objective To obtain adult Trichinella spiralis specific gene sequence. Methods Adult adult cDNA library was screened, cloned, sequenced and compared with GenBank database by DNA probe. DNAstar software was used to analyze the gene sequence. Results The adult cDNA library of adult Trichinella spiralis was screened by subtracted hybridization to obtain a 16 2 9 bp gene. Its open reading frame consists of 146 4 nucleotides and encodes 487 amino acids. Sequence analysis showed that the sequence was an undiscovered Trichinella spiralis gene. There were two zinc finger structures between amino acids 207 ~ 270 in the range of 5 2 ~ 6 4,10 8 ~ 116, 137 ~ 163 , There is a predicted antigenic epitope at amino acids 226 ~ 260. Conclusion Trichinella spiralis adult stage specific genes, the encoded protein has a certain epitope
