目的探讨石衫碱甲在削弱小胶质细胞介导的炎性反应中的作用。方法实验设置有试验对照组、脂多糖作用组、0.1μmol·L~(-1)石衫碱甲预处理组、1μmol·L~(-1)石衫碱甲预处理组、10μmol·L~(-1)石衫碱甲预处理组。MTT试剂检测小胶质细胞的活性,液相芯片技术检测炎性因子的表达。结果实验所用试剂对小胶质细胞活性的影响差异无统计学意义。随着石衫碱甲浓度的增加,脂多糖诱导活化的小胶质细胞释放的IL-6、TNF-α和NO水平也相应显著降低,与脂多糖作用组比较差异有统计学意义(P<0.001)。结论石衫碱甲可削弱小胶质细胞介导的炎性反应。
Objective To investigate the role of strychnine in attenuating the microglial-mediated inflammatory reaction. Methods The experiment was set up with experimental control group, lipopolysaccharide group, 0.1μmol·L -1 rosiglitazone pretreatment group, 1μmol·L -1 rosiglitazone pretreatment group, 10μmol·L ~ (-1) Rosmarin A pretreatment group. MTT reagent to detect microglia activity, liquid chip technology to detect inflammatory factor expression. Results There was no significant difference in the effect of the reagents used on the microglial activity. The levels of IL-6, TNF-α and NO released by lipopolysaccharide-induced microglial cells also decreased significantly with the increase of silibin concentration, which was significantly different from that of lipopolysaccharide group (P < 0.001). Conclusion: Rosmarin can weaken the microglial-mediated inflammatory reaction.
