目的:研究青娥方对去卵巢大鼠骨组织FAK表达及骨代谢生化指标的影响。方法:96只3月龄雌性SD大鼠随机分为假手术组、单纯去卵巢组、骨疏康组、青娥方组。前两组灌服0.9%氯化钠溶液,后两组分别灌服骨疏康及青娥方。分别于用药4、8、12周检测大鼠腰椎FAK m RNA及蛋白含量、胫骨骨密度(BMD)、血清骨碱性磷酸酶(BALP)、酸性磷酸酶(TRAP-5b)、尿脱氧吡啶吩(DPD)、尿肌酐(Cr)等的变化。结果:用药4、8、12周,与假手术组比较,单纯去卵巢组骨FAK m RNA及蛋白表达明显升高(P<0.01),随着时间推移逐渐越低。BMD及血TRAP-5b持续下降,血BALP呈先升高后下降态势,尿DPD/Cr持续升高。青娥方干预4、8、12周,骨FAK m RNA及蛋白表达持续下降,明显低于单纯去卵巢组(P<0.05,P<0.01)。用药8、12周,与单纯去卵巢组比较,青娥方组血BALP、血TRAP-5b水平较之前明显改善(P<0.05,P<0.01);用药12周后,BMD及尿DPD/Cr明显高于单纯去卵巢组(P<0.01),与假手术组相比无明显差异。结论:青娥方能抑制骨组织FAK的表达,提高BMD及血BALP水平,降低血TRAP-5b及尿DPD/Cr含量,降低去卵巢大鼠的骨吸收速度。
Objective: To study the effect of Qing’e prescription on FAK expression and biochemical markers of bone metabolism in ovariectomized rats. Methods: 96 three-month-old female Sprague-Dawley rats were randomly divided into sham operation group, simple ovariectomy group, Gushukang group and Qing’e Fang group. The first two groups were fed with 0.9% sodium chloride solution, and the latter two groups were given Gushukang and Qing’e prescription respectively. The FAK m RNA and protein content in the lumbar spine of rats were measured at 4, 8 and 12 weeks after treatment. BMD, BALP, TRAP-5b and urine deoxypyridine (DPD), urinary creatinine (Cr) and other changes. RESULTS: At 4, 8 and 12 weeks after treatment, FAK m RNA and protein expression in ovariectomized group were significantly higher than those in sham operation group (P <0.01), and gradually decreased with time. BMD and blood TRAP-5b continued to decline, blood BALP first increased and then decreased, urinary DPD / Cr continued to rise. At 4, 8, and 12 weeks after intervention of Qing’E prescription, the expression of FAK m RNA and protein in bone decreased continuously, which was significantly lower than that in simple ovariectomized group (P <0.05, P <0.01). Compared with the simple ovariectomized group, the levels of serum BALP and TRAP-5b in blood in the group of Qing’E prescription were significantly improved at 8 and 12 weeks of treatment (P <0.05, P <0.01) Which was significantly higher than that of the simple ovariectomized group (P <0.01), but no significant difference compared with the sham operation group. CONCLUSION: Qing’e prescription can inhibit the expression of FAK in bone tissue, increase the level of BMD and blood BALP, decrease the content of TRAP-5b and DPD / Cr in urine, and decrease the bone resorption rate in ovariectomized rats.
