以冬小麦农大93(单核期花药)为实验材料,运用 Mg~(2+)-ATP 酶电镜细胞化学定位方法对高温(45±0.5)C 处理(0~30min)后花药的 Mg~(2+)-ATP 酶变化特点观察结果表明:单核期花药中不同部位的 Mg~(2+)-ATP 酶活性表现为绒毡层细胞酶活性>乌氏体和小孢子外壁及其内膜>表皮、内壁和中层。高温处理绒毡层细胞、小孢子质膜和细胞器膜最易破碎,并伴随 ATP 酶活性下降,其下降幅度明显高于其他部位。由此可见,绒毡层细胞和小孢子的 Mg~(2+)-ATP 酶活性及膜结构对高温比花药的表皮、内壁和中层敏感。
The experiment was carried out using electron microscopy of Mg ~ (2 +) - ATPase at the temperature of (45 ± 0.5) C for 0 ~ 30min. +) - ATPase showed that the Mg ~ (2 +) - ATPase activity of different parts of anther in mononuclear stage showed the activity of tapetum> the activity of umbilicus and microspore outer wall and its intima> Epidermis, inner wall and middle layer. High temperature treatment of tapetum cells, microspore membrane and organelle membrane most easily broken, accompanied by decreased ATPase activity, the decrease was significantly higher than the other parts. Thus, Mg ~ (2 +) - ATPase activity and membrane structure of tapetum cells and microspores were sensitive to high temperature than the anther epidermis, inner wall and middle layer of anther.
