目的 为观察含黄连解毒汤血清对肝损伤小鼠巨噬细胞产生IL -6、IL -1的影响。方法 采用灌胃高、中、低三剂量黄连解毒汤 3天后心脏取血制备含黄连解毒汤血清 ,腹腔注射D -氨基半乳糖制作肝损伤动物模型 ,取该小鼠腹腔巨噬细胞 ,在体外加脂多糖、含中药血清 ,收集巨噬细胞内、外液 ,检测IL -6、IL -1水平。结果表明 ,含黄连解毒汤血清高、中、低三剂量均可有效提高巨噬细胞外IL-6水平 ;高剂量可提高巨噬细胞内IL -6水平。用该方法未检测到IL -1。结论 :含黄连解毒汤血清可提高肝损伤小鼠巨噬细胞产生IL -6水平。
Objective To observe the effect of serum containing Huanglian Jiedu Decoction on the production of IL-6 and IL-1 by macrophages in mice with liver injury. METHODS: After three days of high-dose, middle-dose, and low-dose Huanglian Jiedu Decoction, blood was collected from the heart to prepare serum containing Huanglian Jiedu Decoction and an animal model of hepatic injury was established by intraperitoneal injection of D-galactosamine. The mouse peritoneal macrophages were taken in vitro. Lipopolysaccharides, serum containing Chinese herbs, and macrophage internal and external fluids were collected to detect IL-6 and IL-1 levels. The results showed that the high, medium, and low doses of Huanglian Jiedu Decoction can effectively increase the level of macrophage IL-6; high doses can increase IL-6 levels in macrophages. IL-1 was not detected by this method. Conclusion : Serum containing Huanglianjiedu Decoction can increase the level of IL-6 produced by macrophages in mice with liver injury.
