目的:研究大鼠孕期维生素D缺乏对其子鼠骨发育相关基因BMP-2、Cbfa1、Dmp1表达的影响。方法:成熟雌性SD大鼠36只随机分为模型组和对照组各18只,模型组以不含维生素D的大鼠饲料避光喂养,对照组以标准饲料正常光照喂养。两周后与成熟SD雄性大鼠交配,分别取孕20天(E20d)、生后1天(B1d)和生后7天(B7d)子鼠左侧股骨。通过Real-TimePCR方法分别比较模型组和对照组胚胎和新生鼠长骨中BMP-2、Cbfa1、Dmp1基因在3个时间点表达的差异。结果:BMP-2mRNA、Cbfa1mRNA及Dmp1mRNA在E20d、B1d和B7d的表达,模型组均明显低于对照组,差异有统计学意义(P<0.05)。结论:BMP-2、Cbfa1、Dmp1均为骨发育相关基因,孕鼠维生素D缺乏可下调子鼠长骨中BMP-2、Cbfa1、Dmp1的表达水平,从而可能影响子鼠骨骼的发育。
OBJECTIVE: To study the effect of vitamin D deficiency during pregnancy on BMP-2, Cbfa1 and Dmp1 expression in bone development-related genes of their offspring. Methods: Thirty-six adult female Sprague-Dawley rats were randomly divided into model group and control group with 18 rats in each group. The rats in the model group were protected from light by the diet containing vitamin D, while those in the control group were fed normal light. Mice were mated with mature SD male rats two weeks later. Left femur of the offspring were taken at 20 days of gestation (E20d), one day after birth (B1d) and 7 days after birth (B7d). Real-TimePCR methods were used to compare the expression of BMP-2, Cbfa1 and Dmp1 in the long bones of model and control groups at three time points respectively. Results: The expressions of BMP-2 mRNA, Cbfa1 mRNA and Dmp1 mRNA on E20d, B1d and B7d in model group were significantly lower than those in control group (P <0.05). CONCLUSION: BMP-2, Cbfa1 and Dmp1 are both bone development-related genes. Vitamin D deficiency in pregnant rats can down-regulate the expression of BMP-2, Cbfa1 and Dmp1 in the long bones of the offspring, which may affect the development of bone in the offspring.
