次黄嘌呤脱氢酶(inosine 5’-monophosphate dehydrogenase,IMPDH)是GMP从头合成途径的限速酶,多种疾病会影响其活性与表达。以它为靶点的药物已广泛应用到抗病毒、免疫抑制、抗肿瘤等领域。IMPDH不仅充当核苷酸代谢酶的角色,它还具有多种其他生物学功能,在疾病及生理过程中发挥着重要作用。本文就IMPDH的基本功能、催化机制、生物学功能、抑制剂分类及其作为药物靶点的意义和研究进展作一综述,希望为进一步发现IMPDH的生物学功能与新型抑制剂提供参考。
Inosine 5’-monophosphate dehydrogenase (IMPDH) is a rate-limiting enzyme in the de novo synthesis of GMP, and many diseases affect its activity and expression. It targets the drug has been widely used in anti-virus, immunosuppression, anti-tumor and other fields. Not only does IMPDH act as a nucleoside metabolic enzyme, it also has a variety of other biological functions that play an important role in disease and physiology. This review summarizes the basic functions, catalytic mechanisms, biological functions, inhibitor classifications and their significance as drug targets of IMPDH and their research progress. We hope to provide a reference for further discovering the biological functions and new inhibitors of IMPDH.
