目的:探讨通心络超微粉对糖尿病大鼠心肌细胞凋亡的干预作用,为临床用药提供实验依据。方法:取SD大鼠随机分为3组:正常对照组、糖尿病心肌病变组(Diabetic Myocardi-opathy,DCM组)和通心络干预组(TXL组)。采用Tunel法检测光镜下观察心肌细胞凋亡指数,免疫组化法测定Bcl-2,Bax及P53基因蛋白表达。结果:TXL组大鼠较DCM组大鼠心肌细胞凋亡指数明显下降(P<0.01),Bcl-2表达明显增多(P<0.01),Bax及P53表达明显减少(P<0.01)。结论:通心络超微粉可以通过多种途径有效减少糖尿病大鼠心肌细胞凋亡。
Objective: To investigate the intervention effect of Tongxinluo ultrafine powder on myocardial cell apoptosis in diabetic rats and provide experimental basis for clinical drug use. Methods: SD rats were randomly divided into 3 groups: normal control group, Diabetic Myocardi-opathy group (DCM group) and Tongxinluo intervention group (TXL group). The apoptotic index of cardiomyocytes was observed under light microscope by Tunel assay, and the protein expression of Bcl-2, Bax and P53 was measured by immunohistochemistry. RESULTS: Compared with DCM group, the apoptosis index of TXL rats significantly decreased (P<0.01), Bcl-2 expression increased significantly (P<0.01), Bax and P53 expression decreased significantly (P<0.01). Conclusion: Tongxinluo ultrafine powder can effectively reduce cardiomyocyte apoptosis in diabetic rats through various ways.
