目的:探讨类风湿关节炎(RA)患者外周血单核细胞(PBMCs)Toll样受体(TLR)2、4mRNA和蛋白的表达及脂多糖(LPS)对其表达的影响。方法:收集已确诊的RA患者5例(DAS28≥2.6),健康自愿者5例;体外分别分离培养受试者PBMCs作为RA组和健康组,下设空白组与LPS组,其中LPS组给予100μg/L LPS进行刺激,细胞培养24 h后,FQ-RT PCR检测单核细胞TLR2、TLR4 mRNA表达;Western blot检测单核细胞TLR2、TLR4蛋白表达。结果 :给予LPS刺激后,RA组与健康组PBMCs TLR2、TLR4 mRNA及其蛋白的表达均高于相应组刺激前(P<0.001);给予LPS刺激前和刺激后RA组PBMCs TLR2、TLR4 mRNA及其蛋白的表达水平均明显高于健康组(P<0.001)。结论:LPS可刺激RA患者PBMCs已高表达TLR2、TLR4的表达水平更高,可能与机体的炎症反应有关。
Objective: To investigate the expression of TLR 2,4 mRNA and protein in peripheral blood mononuclear cells (PBMCs) and the effect of lipopolysaccharide (LPS) on the expression of rheumatoid arthritis (RA) patients. Methods: Five patients with confirmed RA (DAS28≥2.6) and five healthy volunteers were enrolled in this study. PBMCs were isolated and cultured in vitro as RA group and healthy group, respectively. Blank group and LPS group were established. LPS group received 100μg / L LPS for 24 h. The expression of TLR2 and TLR4 mRNA in monocytes was detected by FQ-RT PCR and the expression of TLR2 and TLR4 in monocytes were detected by Western blot. Results: After LPS stimulation, the expressions of TLR2 and TLR4 mRNA and protein in PBMCs of RA group and healthy group were significantly higher than those of the corresponding groups (P <0.001). Before and after LPS stimulation, The protein expression levels were significantly higher than the healthy group (P <0.001). CONCLUSION: TLR2 is highly expressed in PBMCs of patients with LPS-stimulated RA, and the expression of TLR4 is higher, which may be related to the inflammatory reaction.
