目的研究丹参酮ⅡA(Tan ⅡA)联合顺铂(DDP)在体外对宫颈癌Hela细胞的凋亡作用及凋亡蛋白Bax与Bcl-2表达水平的变化。方法采用苏木素-伊红染色法观察细胞的形态学变化;流式细胞仪检测药物作用后的细胞凋亡情况;Western Blot测定Bax与Bcl-2蛋白在Hela细胞中的表达水平。结果对照组细胞未见凋亡,而两药单用组及联合组均可见凋亡细胞;除对照组外其他各组均可检测到细胞亚二倍体凋亡峰,Tan ⅡA+DDP组凋亡率高于两药单用组凋亡率,差别均有统计学意义(P<0.05);Bcl-2蛋白在各组中均有表达,但表达水平变化不明显(P>0.05);Bax蛋白在对照组中几乎不表达,而在各药物处理组中均有表达,且两药联合组中表达水平高于两药单用组,差别均有统计学意义(P<0.05)。结论Tan ⅡA与DDP二者联合可以增加Hela细胞凋亡率,其中通过增加Bax蛋白的表达,使Bax/Bcl-2比值升高,可能是其诱导细胞凋亡的机制之一。
Objective To investigate the apoptosis of cervical cancer Hela cells and the expression of Bax and Bcl-2 proteins in Tan Ⅱ A combined with cisplatin (DDP) in vitro. Methods Morphological changes of Hela cells were observed by hematoxylin-eosin staining. Apoptosis was detected by flow cytometry. Western Blot was used to detect the expression of Bax and Bcl-2 protein in Hela cells. Results There was no apoptotic cells in the control group, but apoptotic cells were found in the two groups alone and in combination group. In all groups except the control group, (P <0.05). Bcl-2 protein was expressed in all the groups, but the expression level was not changed significantly (P> 0.05); Bax The protein was almost not expressed in the control group, but was expressed in all the drug-treated groups. The expression level of the protein in the two groups was higher than that in the two groups (P <0.05). Conclusions The combination of Tan Ⅱ A and DDP can increase the apoptosis rate of Hela cells. The increase of Bax / Bcl-2 ratio may be one of the mechanisms of apoptosis induction.
