本文推荐一种定量检测血清中免疫复合物(IC)的简易方法——聚乙二醇(PEG)紫外分光比色法,其原理是根据低浓度的PEG在碱性溶液中能沉淀血清中的IC,将沉淀后的IC用NaOH溶解进行紫外分光比色,与凝聚的IgG比较求出IC含量(相当于凝聚IgG微克/毫升)。并对试验的精密度进行了试测,证明有较好的重复性与回收率。实验测得正常人100例血清IC16.72±8.68微克/毫升,各种病例187例,其中以全身性红斑狼疮(SLE)与类风湿性关节炎(RA)最高,平均含量与阳性检出率分别为106.60±22.00微克/毫升(100%)及44.68±15.83微克/毫升(80%)。本法不用超速离心,操作简便,为临床常规定量血清IC提供了一种有用的方法。
This article recommends a simple method for the quantitative detection of immune complexes in serum (IC) - polyethylene glycol (PEG) UV spectrophotometry, the principle is based on the low concentration of PEG in alkaline solution can precipitate serum IC, the precipitated IC dissolved in NaOH for UV spectrophotometry colorimetric, and compared with the aggregated IgG to determine the IC content (equivalent to agglutination IgG micrograms / ml).
