目的 :了解抗癌药物对细胞株增长的影响及凋亡诱导作用。方法 :药物与细胞株共同孵育不同时间后 ,应用四唑盐(MTT)比色法及流式细胞仪 (FCM )分别测定细胞增长抑制率 (CI)及细胞凋亡指数 (APOI)。结果 :抗癌药物组CI值及APOI值与对照组相比均有显著差异 (P <0 .0 5 )。经相关分析得出 ,APOI与CI呈正相关 (r =0 .4 2 ,P <0 .0 5 )。结论 :7种抗癌药物均能明显抑制细胞株增长 ,并可诱导其发生凋亡。利用MTT法及FCM测定细胞CI和APOI,有利于化疗方案的设计 ,指导临床合理用药。
Objective: To understand the effects of anticancer drugs on cell growth and apoptosis induction. Methods: After incubation with drug and cell lines for different time, MTT assay and flow cytometry (FCM) were used to determine the cell growth inhibition rate (CI) and apoptosis index (APOI). Results: The CI and APOI values of anticancer drug group were significantly different from those of the control group (P <0.05). Correlation analysis showed that APOI was positively correlated with CI (r = 0.42, P <0.05). Conclusion: All 7 kinds of anticancer drugs can significantly inhibit cell growth and induce apoptosis. The use of MTT assay and FCM determination of cell CI and APOI, is conducive to the design of chemotherapy regimens, to guide clinical rational use of drugs.
