方法:采用细胞株体外培养技术,研究β-胡萝卜素和亚硒酸钠(Na2SeO3)单独/联合对人肝癌细胞株(Q3)和人膀胱癌细胞株(T24)生长的影响。以小鼠成纤维母细胞(3T3)为正常对照细胞株。实验分为:1.β-胡萝卜素组:剂量为3、30、100μmol/L,溶剂对照为DMSO;2.亚硒酸钠组:剂量为0.15、1.5、5.0μmol/L,溶剂对照为生理盐水;3.β-胡萝卜素和亚硒酸钠联合组:剂量为1.5+0.15、15+1.5、50+5.0μmol/L,溶剂对照为DMSO和生理盐水。各实验组均设空白组。分别在实验后24h、48h、72h观察细胞成活率和活细胞蛋白质含量的变化。结果:1.β-胡萝卜素各剂量组对两株肿瘤细胞生长均有明显的抑制作用;2.亚硒酸钠对两株肿瘤细胞的抑制作用较弱,但随作用时间延长和剂量的增加其抑制作用明显增强;3.β-胡萝卜素和亚硒酸钠对Q3细胞株的抑制作用比T24细胞株强;4.β-胡萝卜素和亚硒酸钠联合对Q3和T24细胞的抑制作用结果提示两者有相加效应。
Methods: The effects of β-carotene and sodium selenite (Na2SeO3) alone or in combination on the growth of human hepatocellular carcinoma cell line (Q3) and human bladder cancer cell line (T24) were studied using cell line in vitro culture. Mouse fibroblasts (3T3) as a normal control cell line. The experiment is divided into: 1. β-carotene group: the dose of 3,30,100μmol / L, solvent control DMSO; Sodium selenite group: the dose of 0.15, 1.5, 5.0μmol / L, solvent control saline; 3. β-carotene and sodium selenite combination group: the dosage of 1.5 + 0.15, 15 + 1.5, 50 + 5.0μmol / L, the solvent control was DMSO and saline. Each experimental group were set blank group. The changes of cell viability and protein content in living cells were observed at 24h, 48h and 72h after the experiment. Results: 1. β-carotene dose of the two groups of tumor cell growth were significantly inhibited; 2. The inhibitory effect of sodium selenite on the two tumor cells is weak, but with the prolonged action and dose increase its inhibitory effect was significantly enhanced; 3. The inhibitory effect of β-carotene and sodium selenite on Q3 cell line was stronger than that of T24 cell line.4. The results of combined inhibition of β-carotene and sodium selenite on Q3 and T24 cells suggest additive effect between the two.
