目的:研究疏肝补肾中药对大鼠乳腺癌癌前病变模型EGFR/STAR3细胞通路的影响,探讨疏肝补肾中药对于阻断大鼠乳腺癌癌前病变的可能机制。方法:SD大鼠二甲基苯恩法造模后分为模型组、消核冲剂低剂量组、消核冲剂高剂量组,每组20只分别中药处理,光镜观察、免疫组化方法检测各组的大鼠模型乳腺组织病理形态、ER、PR表达及EGF、EGFR、STAT3蛋白表达。结果:光镜观察结果显示中药可部分逆转及阻断乳腺癌癌前病变进程;免疫组化方法显示中药组较模型组EGFR、STAR3表达降低(P<0.05),作用随剂量增加而增强。结论:疏肝补肾中药可以通过阻断EGFR/STAR3细胞信号传导通路的途径抑制雌激素分泌从而达到逆转阻断乳腺非典型增生发展的作用,使其恢复正常状态,并且作用程度随着剂量的增加而增加。
Objective: To study the effect of Shugan Bushen Chinese herbs on EGFR / STAR3 cell pathways in rat model of precancerous lesion of the breast and to explore the possible mechanism of Shugan Bushen Chinese herbs in blocking precancerous lesions of rat breast cancer. Methods: SD rats were divided into model group, low dosage group for extinction granules and high dosage group for extinction granules, 20 rats in each group were treated with traditional Chinese medicine, light microscopy and immunohistochemistry The histopathology, expression of ER, PR and the expression of EGF, EGFR and STAT3 in the rat model of each group. Results: The results of light microscopy showed that Chinese herbs could partially reverse and block the progression of precancerous lesions of breast cancer. Immunohistochemistry showed that the expressions of EGFR and STAR3 were lower in Chinese medicine group than those in model group (P <0.05). Conclusion: Shugan Bushen Chinese herbs can inhibit the secretion of estrogen by blocking the signal pathway of EGFR / STAR3 cells so as to reverse the effect of blocking the development of atypical mammary gland hyperplasia and restore normal estrogen status. The effect of Shugan Bushen Chinese herbs can be reversed with dose increasing Increase.
