目的探讨白术对“泻剂结肠”的治疗作用及其可能机制。方法健康成年SPF级SD大鼠30只,采用“大黄酸混悬液灌胃法”制作“泻剂结肠”大鼠动物模型。制模成功后,随机分为生理盐水组(6只)、白术组1(6只)、莫沙必利组1(6只)、白术组2(6只)、莫沙必利组2(6只)。生理盐水组以生理盐水灌胃30天;白术组1以白术灌胃30天;白术组2以白术灌胃30天后,再正常饲养30天;莫沙必利组1以莫沙必利灌胃30天;莫沙必利组2以莫沙必利灌胃30天后,再正常饲养30天。检测首粒黑便排出时间及粪便干湿重比以反映肠道传输功能及粪便性质。应用酶联免疫分析法检测大鼠血清中VIP的含量。结果与生理盐水组相比,白术组1首粒黑便排出时间明显缩短,粪便干湿重比明显降低,血清VIP含量明显下降,均差别显著(P<0.05)。与莫沙必利组2比较,白术组2首粒黑便排出时间明显缩短,粪便干湿重比明显降低,有统计学意义(P<0.05)。结论白术能够显著改善“泻剂结肠”大鼠的肠道传输功能及粪便干湿重比,远期疗效优于莫沙必利。白术治疗“泻剂结肠”的机制可能与其降低血清中VIP含量有关。
Objective To investigate the therapeutic effect of Atractylodes on “laxative colon” and its possible mechanism. Methods Thirty healthy adult Sprague-Dawley (SD) SD rats were treated with “rhein suspension by gavage ” to make “Laxative Colon” rat model. After successful modeling, the rats were randomly divided into saline group (n = 6), Atractylodes group (n = 6), mosapride group (n = 6), Atractylodes group (n = 6), mosapride group 2 6). The rats in the normal saline group were treated with saline for 30 days. The rats in the atractylodes rhizome of Baiji operation group were treated with atractylodes rhizome for 30 days. 30 days; mosapride group 2 to mosapride gavage after 30 days, and then normal for 30 days. Detection of the first black discharge time and dry weight ratio of wet to reflect the intestinal transit function and the nature of the feces. Enzyme-linked immunosorbent assay was used to detect the content of VIP in serum of rats. Results Compared with the saline group, the discharge time of the first granule in the atractylodes rhizome of Atractylodes macrocephalae group was significantly shortened, the ratio of wet to dry weight of stool was significantly decreased, and the serum VIP level was significantly decreased (P <0.05). Compared with mosapride group 2, the discharge time of the first black tablets in the atractylodes rhizome group was significantly shortened and the ratio of wet to dry weight of feces was significantly decreased (P <0.05). Conclusion Atractylodes macrocephalae can significantly improve the intestinal transit function and the ratio of dry to wet weight of stool in rats with cathartic laxatives, and the long-term curative effect is better than that of mosapride. Atractylodes the mechanism of “cathartic colon” may be related to its lower serum VIP content.
