用高效液相色谱(HPLC)法测定大鼠衰老过程中肝细胞活性染色质与非活性染色质DNA甲基化程度的变化。结果表明,老年鼠肝细胞活性染色质DNA甲基化程度较青年鼠的低,二者有显著性差异;而老年鼠肝细胞非活性染色质DNA甲基化程度较青年鼠高,亦具有显著性差异,这表明衰老过程中基因组DNA各组分甲基化程度的变化不一致。
The level of methylation of active and non - active chromatin in rat hepatocytes during aging was determined by high performance liquid chromatography (HPLC). The results showed that the degree of methylation of activated chromatin DNA in aged rat hepatocytes was lower than that in young mice, and the difference was statistically significant. However, the methylation of non-active chromatin DNA in aged rats was higher than that in young mice This indicates that there is inconsistent change of the methylation level of genomic DNA during the aging process.
