目的:研究痰热清(TRQ)对于内毒素性急性肺损伤(ALI)兔的肺局部多形核细胞(PMN)凋亡影响及其抗氧化作用。方法:50只健康兔随机分组(n=10),静脉注射内毒素建立急性肺损伤模型(模型组),应用TRQ(高中低剂量3组)预处理后,检测肺通透指数及病理变化,并用流式细胞仪测定外周血及灌洗液内的PMN凋亡、坏死细胞比例及呼吸爆发功能,肺组织匀浆中超氧化物歧化酶(SOD)、脂质过氧化产物丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)水平变化。结果:内毒素诱导的兔ALI中,ALI模型组MDA含量显著多于对照组,SOD活性和GSH-Px含量都明显低于对照组(P<0.01),BALF中PMN成活比例增加,凋亡延迟,呼吸爆发明显增强。而TRQ高中低剂量的使用均可抑制上述各变化(P<0.05)并明显改善肺组织病理变化。结论:TRQ的抗氧化作用可阻止内毒素诱导的兔ALI的发生与发展,对其有明显的保护作用。
OBJECTIVE: To investigate the effect of Tanreqing (TRQ) on apoptosis of pulmonary local polymorphonuclear cells (PMN) in rabbits with endotoxin-induced acute lung injury (ALI) and its antioxidative effect. METHODS: Fifty healthy rabbits were randomly divided into groups (n=10). Acute lung injury model (model group) was established by intravenous injection of endotoxin. Lung permeability index and pathological changes were measured after TRQ (high, low dose 3) pretreatment. Flow cytometry was used to measure PMN apoptosis, necrotic cell ratio and respiratory burst in peripheral blood and lavage fluid. Superoxide dismutase (SOD) and lipid peroxidation product malondialdehyde (MDA) in lung homogenate were measured. Changes in glutathione peroxidase (GSH-Px) levels. RESULTS: Endotoxin-induced ALI in rabbits was significantly more MDA content in the ALI model group than in the control group. SOD activity and GSH-Px content were significantly lower than those in the control group (P<0.01). The survival rate of PMN in the BALF was increased and the apoptosis was delayed. The respiratory outburst significantly increased. The use of TRQ high, medium and low doses inhibited the above changes (P<0.05) and significantly improved lung pathological changes. Conclusion: The antioxidant effect of TRQ can prevent the occurrence and development of ALI induced by endotoxin in rabbits, and it has obvious protective effect on it.
