目的建立塔里木盆地17个引进红枣品种高效液相色谱指纹图谱方法,为红枣品质的科学评价和产品的质量控制提供科学依据。方法采用HPLC法,色谱柱TOSOH TSKgel ODS-100V C18柱(150 mm×4.6 mm,3μm),流动相为甲醇-0.3%磷酸二氢钾缓冲水溶液,梯度洗脱,体积流量1.0 m L/min,柱温40℃,检测波长260 nm。对采自塔里木大学种质资源圃中17个引进品种的红枣进行指纹图谱研究,用药典委员会颁布的中药色谱指纹图谱相似度评价系统2004A进行评价。结果建立了红枣的指纹图谱,标定了22个共有峰,各峰分离度良好,并对尿嘧啶、次黄嘌呤、腺嘌呤、胞苷、尿苷、环磷酸腺苷(c AMP)、环磷酸鸟苷(c GMP)进行测定。结论 HPLC指纹图谱分析技术可用于红枣的鉴别。
Objective To establish a method for the identification of 17 jujube cultivars in Tarim Basin by high performance liquid chromatography (HPLC) fingerprinting, which provides a scientific basis for scientific evaluation of jujube quality and product quality control. Methods The mobile phase consisted of methanol-0.3% potassium dihydrogen phosphate buffer solution (gradient elution) at a flow rate of 1.0 mL / min on a TOSOH TSKgel ODS-100V C18 column (150 mm × 4.6 mm, Column temperature 40 ℃, detection wavelength 260 nm. The fingerprinting of jujube from 17 varieties introduced from Tarim University germplasm resource nursery was conducted and evaluated by the similarity evaluation system 2004A of TCM chromatographic fingerprint promulgated by Pharmacopoeia Commission. Results Fingerprints of jujube were established and 22 common peaks were calibrated. The peak separation was good. The uracil, hypoxanthine, adenine, cytidine, uridine, cAMP, Guanosine (c GMP) was determined. Conclusion HPLC fingerprinting can be used to distinguish jujube.
