目的阐明苓桂术甘汤合葶苈大枣泻肺汤治疗慢性心衰(CHF)的作用机制。方法 Wistar大鼠随机分为空白对照组(K组)和A组,A组用阿霉素(Adr)腹腔注射制备CHF模型。6周造模结束后,再将A组大鼠随机分为模型对照组(M组)、温阳消饮组(W组)、去桂消饮组(Q组)。除M组、K组外,W组、Q组大鼠在末次注射Adr后2 h分别予以相应药物灌胃,日1次,连续4周。然后检测各组大鼠心房利钠肽(ANP)、脑钠肽(BNP)、血管紧张素Ⅱ(AngⅡ)。结果与K组比较,M组的ANP显著升高(P<0.01);与M组比较,W组、Q组的ANP水平有差异,但无统计学意义(P>0.05);与K组比较,M组的BNP显著升高(P<0.01);与M组比较,W组、Q组的BNP水平显著降低(P<0.01);与K组比较,M组的AngⅡ显著升高(P<0.05);与M组比较,W组、Q组的AngⅡ水平明显降低(P<0.05)。结论苓桂术甘汤合葶苈大枣泻肺汤能提高CHF大鼠血清BNP、AngⅡ含量,这可能是其治疗CHF的作用机制之一。
Objective To elucidate the mechanism of action of Lingguizhugan Decoction combined with Jujube Xiefei Decoction in treating chronic heart failure (CHF). Methods Wistar rats were randomly divided into blank control group (group K) and group A, and group A was injected intraperitoneally with adriamycin (Adr) to establish CHF model. After 6 weeks of modeling, the rats in group A were randomly divided into model control group (M group), Wenyangxiaoying group (W group) and QGG group (Q group). Except M group and K group, rats in W group and Q group were given gavage 2 h after the last injection of Adr respectively, once a day for 4 weeks. Then atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP) and angiotensin Ⅱ (AngⅡ) were detected in each group. Results Compared with K group, the ANP level in M group was significantly increased (P <0.01). Compared with M group, the levels of ANP in W group and Q group were not significantly different (P> 0.05); Compared with K group (P <0.01). Compared with M group, BNP levels in W group and Q group were significantly decreased (P <0.01). Compared with K group, Ang Ⅱ in M group was significantly increased (P < 0.05). Compared with M group, the levels of AngⅡ in W group and Q group were significantly decreased (P <0.05). Conclusions Lingguizhugan decoction combined with Jujube Xiefei Tang can increase serum BNP and AngⅡ levels in CHF rats, which may be one of the mechanisms of CHF treatment.
