为探讨肿瘤坏死因子-α(TNF-α)与急性白血病(AL)的关系,采用双抗体夹心酶联免疫吸附试验法(ELISA)对65例不同病期AL患儿血清TNF-α水平进行检测。结果显示:血清TNF-α水平与正常对照组(7.32+4.31ng/ml)比较,初诊时急性期组(12.01±4.40ng/ml)及复发组(12.59±2.79ng/ml)明显增高(P<0.01),完全缓解组(7.73±4.17ng/ml)则无明显差异(P>0.05)。对急性期组T淋巴细胞亚群检测显示,CD4~+降低,CD8~+增高,CD4~+/CD8~+比值降低。提示血清TNF-α水平可作为AL患儿疾病活动性、病情监测及疗效判断的指标之一,患儿免疫功能紊乱可能与发病有关。
To investigate the relationship between tumor necrosis factor-α (TNF-α) and acute leukemia (AL), serum levels of TNF-α in 65 children with AL at different stages were detected by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) . The results showed that the level of serum TNF-α was significantly higher in the acute phase group (12.01 ± 4.40ng / ml) and the recurrence group (12.59 ± 2.79ng / ml) compared with the normal control group (7.32 ± 4.31ng / ml) <0.01). There was no significant difference in complete remission group (7.73 ± 4.17ng / ml) (P> 0.05). The detection of T lymphocyte subsets in acute stage showed that CD4 ~ + decreased, CD8 ~ + increased, CD4 ~ + / CD8 ~ + ratio decreased. It is suggested that the level of serum TNF-α may be used as an indicator of disease activity, disease monitoring and curative effect judgment in AL children. The immune dysfunction in children may be related to the pathogenesis.
