本文建立了测定血浆中吗啡浓度的气相色谱-质谱联用检测法。以纳洛酚做为内标物,吗啡和内标分离良好,保留时间分别为:吗啡4.8min,内标6.2min。吗啡的相对回收率为79-83%,日内精密度为1.40-2.26%,日间精密度为0.59-1.84%,最低检测浓度为2ng/ml。多种药物均不影响吗啡的测定。本文用此法测定了正常人单次口服吗啡的血药浓度证明本方法是灵敏,专一,准确可靠的,完全能满足人体内吗啡药代动力学研究的要求。
In this paper, a gas chromatography-mass spectrometry method for the determination of morphine in plasma has been established. With nalorphin as an internal standard substance, morphine and internal standard were well separated with retention times of 4.8 minutes for morphine and 6.2 minutes for internal standard. The relative recovery of morphine was 79-83%, the intra-day precision was 1.40-2.26%, the intra-day precision was 0.59-1.84% and the lowest detection concentration was 2ng / ml. A variety of drugs do not affect the determination of morphine. This method was used to determine the normal plasma concentration of a single oral morphine proves that the method is sensitive, specific, accurate and reliable, fully able to meet the human body morphine pharmacokinetic study requirements.
