目的 探讨电磁辐射急性辐照对PC12细胞的影响。方法 对离体培养的PC12细胞以65mW /cm2 电磁波急性照射2 0min ,于辐照后即刻、3、12、2 4h四个时相点检测细胞存活率、乳酸脱氢酶释放率,并采用Annexin -V -FITC及PI双标记流式细胞仪检测PC12细胞凋亡率。结果 电磁波辐照后即刻PC12细胞存活率和LDH释放率变化不明显(P >0 . 0 5 ) ,辐照后3h细胞存活率明显降低,而LDH释放率明显增加(P <0 .0 5 ) ,到2 4h后达到峰值(P <0 . 0 1)。电磁波辐照后即刻PC12细胞凋亡开始增加(P <0 . 0 5 ) ,3h后凋亡细胞进一步明显增多,凋亡率约为2 0 % ,2 4h后细胞凋亡再次显著增加(P <0 . 0 1)。结论 电磁辐射急性辐照后早期即可引起PC12细胞损伤,而在辐照后2 4h出现一种“继发损伤反应”,细胞凋亡可能是电磁辐射辐照诱导PC12细胞损伤的主要原因之一。
Objective To investigate the effect of acute radiation of electromagnetic radiation on PC12 cells. Methods PC12 cells cultured in vitro were irradiated with 65 mW / cm2 electromagnetic waves for 20 min. The cell viability and lactate dehydrogenase release rate were measured at 4, 3, 12 and 24 h after irradiation. Annexin -V-FITC and PI double labeling flow cytometry PC12 cell apoptosis rate. The apoptosis of PC12 cells began to increase immediately after irradiation with electromagnetic wave (P <0.05). After 3 hours, the number of apoptotic cells increased further and the apoptosis rate was about 20%. The apoptosis increased again after 24 hours (P < 0. 0 1). Conclusion Electromagnetic radiation can induce PC12 cell injury in the early stage after acute radiation, and a “secondary damage reaction” occurs 24 h after irradiation. Apoptosis may be one of the main causes of PC12 cell damage induced by electromagnetic radiation .
