目的观察重组白细胞介素13对3T3成纤维细胞巨噬细胞炎症蛋白-1αmRNA表达的影响。方法分别用DMEM(对照组)和40ng/ml、80ng/ml重组白细胞介素13(干预组)培养液作用于3T3成纤维细胞,24h和48h后MTT法观察细胞生长情况,用细胞增殖率表示;两组细胞24h后原位杂交方法检测巨噬细胞炎症蛋白-1αmRNA的表达。结果重组白细胞介素13可显著促进3T3成纤维细胞增殖,且呈时间-剂量效应关系(P<0.01)。对照组及重组白细胞介素13组均可检测到巨噬细胞炎症蛋白-1αmRNA的表达,但与对照组相比,重组白细胞介素13显著促进巨噬细胞炎症蛋白-1αmRNA的表达。结论重组白细胞介素13促进成纤维细胞分泌巨噬细胞炎症蛋白-1α,可能在肺纤维化的发生、发展过程中起重要作用。
Objective To observe the effect of recombinant interleukin-13 on the mRNA expression of macrophage inflammatory protein-1α in 3T3 fibroblasts. Methods 3T3 fibroblasts were treated with DMEM (control group) and 40ng / ml and 80ng / ml recombinant interleukin-13 (intervention group) respectively. The cell growth was observed by MTT method at 24h and 48h, The expression of inflammatory protein-1αmRNA in macrophages was detected by in situ hybridization in both groups after 24h. Results Recombinant interleukin - 13 could significantly promote the proliferation of 3T3 fibroblasts in a time - dose dependent manner (P <0.01). The expression of macrophage inflammatory protein-1αmRNA was detected in the control group and recombinant interleukin-13 group, but recombinant interleukin-13 significantly enhanced the expression of macrophage inflammatory protein-1αmRNA compared with the control group. Conclusion Recombinant interleukin-13 can promote the secretion of macrophage inflammatory protein-1α by fibroblasts, which may play an important role in the occurrence and development of pulmonary fibrosis.
