目的对尖锐湿疣中人乳头瘤病毒(HPV)cp6108进行序列研究。方法应用TA克隆后测序和PCR产物直接测序进行HPV cp6108的基因序列测定。结果 5例HPV cp6108阳性的尖锐湿疣的模板DNA经TA克隆后测序和PCR产物直接测序结果显示,两种测序结果一致,BLAST比对结果显示,与基因库中HPV cp6108提交序列有99%的一致性,排在比对结果第1位,所有标本的cp6108的序列与网上提交序列对比后发现均有3处相同的密码子发生改变(以起始密码子GCA之G为第1位):第210位核苷酸G(鸟嘌呤,)突变为A(腺嘌呤),导致第70位密码子GAG→GAA,发生同义突变;第229位核苷酸G(鸟嘌呤)突变为A(腺嘌呤),导致第77位密码子GAT→AAT,编码的氨基酸由天冬氨酸(Asp,D)变为天冬酰氨(Asn,N),即发生D77N错义突变;第346位核苷酸A(腺嘌呤)突变为C(胞嘧啶),导致第116位ACT→CCT,编码的氨基酸由苏氨酸(Thr,T)变为脯氨酸(Pro,P),即发生T116P错义突变。结论与基因库提交序列相比,本研究检测到的HPV cp6108序列中有3处突变,其中G70A为同义突变,D77N和T116P为错义突变。
Objective To study the sequence of human papillomavirus (HPV) cp6108 in condyloma acuminatum. Methods The sequence of HPV cp6108 gene was determined by TA cloning sequencing and PCR product sequencing. Results The results of BLAST and TAX sequencing showed that the two DNA sequencing results were consistent. The results of BLAST analysis showed that there was a 99% agreement with the sequence of HPV cp6108 in the GenBank The results showed that all of the cp6108 sequences of all the samples were found to have the same codons changed at the same position (the G of the start codon GCA is the first digit) after comparison with the online submission sequence: Mutation of nucleotide G (guanine) at position 210 to A (adenine) led to the synonymous mutation at GAG → GAA at codon 70. Mutation of the nucleotide at nucleotide position 229 (guanine) to A (adenine) Purine), resulting in the mutation of aspartate (Asn, N) from aspartic acid (Asp, D) to the amino acid at codon 77 of GAT → AAT. The mutation of acid A (adenine) to C (cytosine) resulted in the 116 → ACT → CCT mutation of the encoded amino acid from threonine (Thr, T) to proline (Pro, P) mutation. Conclusion There are 3 mutations in HPV cp6108 sequence detected in this study, including G70A synonymous mutation and D77N and T116P as missense mutation.
