目的:观察定喘汤高、中、低剂量组对呼吸道合胞病毒感染大鼠T细胞亚群及NK细胞活性的影响,探讨不同剂量定喘汤的免疫调节作用。方法:将Wistar大鼠随机分为正常组、模型组、定喘汤高、中、低剂量组各9只,动物造模后正常组和模型组均以等体积的生理盐水灌胃,各中药治疗组分别予定喘汤高、中、低剂量灌胃给药。采用流式细胞术检测各组大鼠外周血中T淋巴细胞亚群及NK细胞活性。结果:与正常组比较,模型组除CD4~+/CD8~+比值升高外,其余各项指标均有所下降,其中CD8~+细胞、CD4~+/CD8~+、CD3-CD161~+细胞的百分比具有显著性差异(P<0.05);与模型组比较,各治疗组除CD4~+/CD8~+比值下降外,其余各项指标均有所升高,且高剂量组各项指标均具有显著性差异(P<0.05),中剂量组的CD4~+/CD8~+、CD3-CD161~+细胞百分比具有统计学意义(P<0.05);高剂量组各项指标明显高于其他治疗组,尤其对于低剂量组而言具有统计学差异(P<0.05)。结论:呼吸道合胞病毒感染Wistar大鼠后使机体的免疫功能紊乱,定喘汤对其具有较好的免疫调节作用。
Objective: To observe the effect of Dingchutang high, medium and low dose groups on the T lymphocyte subsets and NK cell activity of respiratory syncytial virus infected rats and to explore the immune regulation effect of Dingchuan decoction with different doses. Methods: Wistar rats were randomly divided into normal group, model group and Dingchu Tang high, medium and low dose groups, each with 9 rats. Normal animals and model group were given normal saline by intragastric administration. Treatment groups were given asthma soup high, medium and low dose intragastric administration. Flow cytometry was used to detect T lymphocyte subsets and NK cell activity in peripheral blood of rats in each group. Results: Compared with the normal group, the indexes of CD4 + / CD8 + increased in model group except for the increase of CD4 + / CD8 + (P <0.05). Compared with the model group, all the indexes except the CD4 ~ + / CD8 ~ + ratio decreased in all treatment groups, and the indexes of high dose group (P <0.05). The percentage of CD4 ~ + / CD8 ~ + and CD3 ~ CD161 ~ + cells in the middle dose group was significantly higher than that in the high dose group (P <0.05) The treatment group, especially for the low-dose group with a statistically significant difference (P <0.05). Conclusion: Respiratory syncytial virus infection in Wistar rats after the body’s immune dysfunction, Dingchutang on it has a good immune regulation.
