目的研究mesothelin(间皮素,MESO)在上皮性卵巢癌中的表达及其与卵巢癌发生、发展之间的关系。方法采用半定量RT-PCR技术及免疫组化方法分别检测上皮性卵巢癌,卵巢肿瘤和正常卵巢组织中MESO mRNA及其蛋白的表达情况,并分析其表达在不同临床分期和病理分级之间的关系。结果上皮性卵巢癌中MESO mRNA(1.4005±0.4646)及蛋白(2.7857±2.2712)表达水平明显高于良性卵巢肿瘤(0.6463±0.2419,1.2500±1.6125)和正常卵巢组织(0.6439±0.2729,0.9167±1.2401),差异有显著性(P<0.05);MESO mRNA及蛋白在浆液性卵巢癌(1.5255±0.4151,3.3036±2.6141)和子宫内膜样癌(1.5250±0.5419,3.0000±2.3094)中的表达高于其在粘液样癌(1.0675±0.3149,1.0566±1.9242)中的表达,差异有统计学意义(P<0.05);MESO在临床Ⅲ、Ⅳ期卵巢癌中的表达显著高于其在Ⅰ、Ⅱ期卵巢癌中的表达,差异有统计学意义(P<0.05);MESO的表达水平与病理分级、病理类型有关,差异有显著性(P<0.05);MESO的表达水平与患者有无腹水有关(P<0.05),而与患者的年龄、血清CA125水平无关(P>0.05)。结论上皮性卵巢癌MESO mRNA及蛋白的表达增高,MESO可能参与卵巢癌的细胞识别、黏附、转移,并促进其腹腔播散与种植。
Objective To investigate the expression of mesothelin (mesothelin, MESO) in epithelial ovarian cancer and its relationship with the occurrence and development of ovarian cancer. Methods The expression of MESO mRNA and protein in epithelial ovarian cancer, ovarian tumor and normal ovarian tissue were detected by semi-quantitative RT-PCR and immunohistochemistry respectively. The expression of MESO mRNA and protein was analyzed between different clinical stages and pathological grades relationship. Results The expression of MESO mRNA (1.4005 ± 0.4646) and protein (2.7857 ± 2.2712) in epithelial ovarian cancer was significantly higher than that in benign ovarian tumor (0.6463 ± 0.2419, 1.2500 ± 1.6125) and normal ovarian tissue (0.6439 ± 0.2729, 0.9167 ± 1.2401) (P <0.05). The expression of MESO mRNA and protein in serous ovarian cancer (1.5255 ± 0.4151,3.3036 ± 2.6141) and endometrioid carcinoma (1.5250 ± 0.5419,3.0000 ± 2.3094) was higher than that (1.0675 ± 0.3149,1.0566 ± 1.9242), the difference was statistically significant (P <0.05). The expression of MESO in stage Ⅲ and Ⅳ ovarian cancer was significantly higher than that in stage Ⅰ and Ⅱ ovary (P <0.05). The expression of MESO was correlated with pathological grade and pathological type, the difference was significant (P <0.05). The expression of MESO was correlated with the presence or absence of ascites <0.05), but not with age of patients and serum CA125 level (P> 0.05). Conclusions The expression of MESO mRNA and protein in epithelial ovarian cancer is increased. MESO may be involved in the cell recognition, adhesion and metastasis of ovarian cancer, and promote the dissemination and implantation of the tumor.
