目的 :探讨化疗药诱导颊癌细胞凋亡及其相关机制。方法 :选择临床行术前化疗患者的颊癌组织进行原位DNA末端标记 (TUNEL法 )显示凋亡细胞以及免疫组化SP法检测Fas蛋白的表达。结果 :凋亡细胞及凋亡小体散在分布于颊癌组织中 ,化疗后颊癌凋亡细胞发生率增加 ,凋亡指数 (AI)均值为 0 .91,明显高于化疗前AI( 0 .38) (P <0 .0 1) ;与化疗前相比 ,化疗后颊癌细胞Fas表达强度增加 (P <0 .0 1)。结论 :细胞凋亡途径可能是化疗药杀灭颊癌细胞的重要机制 ,Fas系统在化疗药诱导颊癌细胞凋亡发生中发挥关键性作用。由此 ,应用凋亡诱导分子FasL有可能成为一种新的抗癌方法。
Objective: To investigate the chemotherapy-induced apoptosis of buccal carcinoma cells and its related mechanisms. Methods: The apoptotic cells were detected by TUNEL method and the expression of Fas protein was detected by immunohistochemical SP method. Results: Apoptotic cells and apoptotic bodies were scattered in buccal carcinoma tissues. The incidence of apoptotic cells in cheek carcinoma increased after chemotherapy. The mean value of apoptosis index (AI) was 0.91, which was significantly higher than that before chemotherapy (P <0.05). 38) (P <0.01). Compared with those before chemotherapy, the expression of Fas in buccal carcinoma cells increased after chemotherapy (P <0.01). CONCLUSION: The apoptotic pathway may be an important mechanism of chemotherapeutic drugs in killing buccal carcinoma cells. Fas system plays a key role in the apoptosis induced by chemotherapy drugs in buccal carcinoma cells. Thus, the application of apoptosis inducing molecule FasL may become a new anti-cancer method.
