目的:探讨CD68抗体标记的肿瘤相关巨噬细胞TAMs和基质溶解素MMP-7在喉鳞状细胞癌组织的表达及与临床病理参数的关系,探讨两种分子标志物表达与喉癌组织MVD的关系。方法:使用免疫组织化学法检测标本65例(喉鳞状细胞癌组织45例,瘤旁非肿瘤组织20例)中CD68及MMP-7、CD34抗体标记的MVD的表达。结果:CD68阳性表达率,鳞癌组织(82.2%,37/45)与瘤旁组织(15.0%,3/20)比较,差异有统计学意义(P<0.05)。MMP-7阳性率鳞癌组织(71.1%,32/45)与瘤旁组织(25.0%,5/20)比较,差异有统计学意义(P<0.05)。CD34-MVD鳞癌组织(26.52±6.36)高于瘤旁组织(12.23±4.01)。有淋巴结转移组CD68、MMP-7阳性率高于无淋巴结转移组;MMP-7、CD68表达与分化程度无关;MMP-7与肿瘤分期无关,而CD68与肿瘤分期相关;CD68、MMP-7与CD34-MVD有正相关性。结论:两种分子标志物在喉癌组织的高表达及与MVD的正相关性说明了TAMs与MMP-7在促进喉鳞状细胞癌组织的转移及新生血管形成中发挥了作用,可作为判断喉癌侵袭及转移的重要标志物。
Objective: To investigate the expression of CD68 antibody-associated tumor-associated macrophages (TAMs) and matrix metalloproteinase-7 (MMP-7) in laryngeal squamous cell carcinoma and its relationship with clinicopathological parameters. To investigate the relationship between the expression of two molecular markers and MVD relationship. Methods: Immunohistochemistry was used to detect the expression of CD68, MMP-7 and CD34-MVD in 65 cases (45 cases of laryngeal squamous cell carcinoma and 20 cases of non-tumor tissue). Results: The positive rate of CD68 was significantly higher in squamous cell carcinoma (82.2%, 37/45) than in para-tumor tissues (15.0%, 3/20) (P <0.05). The positive rate of MMP-7 in squamous cell carcinoma (71.1%, 32/45) was significantly different from that in adjacent tissues (25.0%, 5/20) (P <0.05). CD34-MVD in squamous cell carcinoma (26.52 ± 6.36) was higher than that in peritumoral tissue (12.23 ± 4.01). The positive rate of CD68 and MMP-7 in lymph node metastasis group was higher than that in non-lymph node metastasis group. The expression of MMP-7 and CD68 was not related to the degree of differentiation. The expression of CD68, MMP-7 and CD68 was not correlated with tumor stage, CD34-MVD has a positive correlation. Conclusion: The high expression of two molecular markers in laryngeal squamous cell carcinoma and its positive correlation with MVD demonstrated that TAMs and MMP-7 play roles in promoting laryngeal squamous cell carcinoma metastasis and neovascularization and can be used as a judgment Laryngeal cancer invasion and metastasis of important markers.
