目的分离纯化退黄藤中的东莨菪内酯并测定其含量。方法采用硅胶柱色谱分离纯化退黄藤中的东莨菪内酯,并根据理化性质及光谱数据鉴定其结构;采用HPLC法测定其含量,色谱柱为Zorbax Eclipse XDB-C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%冰乙酸(39∶61),流速1.0 m L·min~(-1),检测波长345 nm。结果东莨菪内酯3.11~31.1μg·m L~(-1)与峰面积呈良好的线性关系(r=0.9999),平均回收率为99.29%,RSD=1.40%(n=6)。结论所用方法简便、准确、快速,可用于退黄藤药材的质量控制。
OBJECTIVE: To isolate and purify scopoletin from Artemisia annua and determine its content. METHODS: Scopolactone was isolated and purified by silica gel column chromatography and its structure was elucidated on the basis of physico-chemical properties and spectral data. The content was determined by HPLC. The column was Zorbax Eclipse XDB-C 18 column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of methanol - 0.1% glacial acetic acid (39:61) and the flow rate was 1.0 m L · min -1. The detection wavelength was 345 nm. Results Scopolactone showed a good linear relationship (r = 0.9999) with the average recovery of 99.29% and RSD = 1.40% (n = 6) in the range of 3.11-31.1 μg · m L -1. Conclusion The method is simple, accurate and rapid, which can be used to control the quality of medicinal materials of rehmannia.
