目的:探讨survivin和hsa-miR-542-3p在口腔鳞状细胞癌(OSCC)中的表达和相关性。方法 :检测30例OSCC肿瘤组织及癌旁组织中survivin和hsa-mi R-542-3p的表达水平,在OSCC细胞系中验证hsa-miR-542-3p对survivin表达的调控作用;检测30例OSCC患者和30例正常成人血浆中hsa-miR-542-3p的表达水平。采用SPSS17.0软件包对数据进行统计学处理。结果 :OSCC样本中survivin表达上升,而hsa-miR-542-3p表达下降,两者呈负相关。在SCC-15细胞系中,hsa-miR-542-3p对survivin的mRNA和蛋白水平起负调控作用。OSCC患者血浆中hsa-miR-542-3p比正常人显著下降,ROC曲线分析显示其AUC=0.82。结论 :在OSCC中,hsa-miR-542-3p降低与survivin上升相关。血浆hsa-miR-542-3p是潜在的OSCC诊断标志物。
Objective: To investigate the expression and correlation of survivin and hsa-miR-542-3p in oral squamous cell carcinoma (OSCC). Methods: The expression levels of survivin and hsa-mi R-542-3p in 30 OSCC tissues and adjacent tissues were detected. The regulatory effect of hsa-miR-542-3p on survivin expression was tested in OSCC cell lines. OSCC patients and 30 normal adult plasma hsa-miR-542-3p expression levels. SPSS17.0 software package for statistical analysis of the data. Results: The expression of survivin in OSCC samples was increased, while the expression of hsa-miR-542-3p was decreased. There was a negative correlation between them. In SCC-15 cell line, hsa-miR-542-3p negatively regulates survivin mRNA and protein levels. The level of hsa-miR-542-3p in the plasma of patients with OSCC was significantly lower than that of the normal subjects. The ROC curve analysis showed that the AUC was 0.82. Conclusion: In OSCC, the decrease of hsa-miR-542-3p is correlated with the increase of survivin. Plasma hsa-miR-542-3p is a potential diagnostic marker for OSCC.
