本文采用多聚赖氨酸阳离子膜制备人类血细胞的扫描电镜观察样品。由于血细胞的多阴离子表面和多聚赖氨酸的多阳离子表面之间的相互作用,各种细胞可以牢固地吸附到用多聚赖氯酸(Poly-L-lysine)预先处理过的玻璃片上。当细胞与表面接触时,则立即发生细胞的附着。被吸附的细胞伸展好,单层排列,分布均匀,画面清晰,便于观察而不失去细胞原有的鉴别性形态特征。被吸附的生物材料不因制备样品过程中的固定、脱水和临界点干燥等处理而脱落。应用该法制备了人外周血淋巴细胞,建系的淋巴细胞的扫描电镜的观察样品。这些细胞都显示了清晰而立体感强的扫描图象。这对研究各种血细胞及其他的游离细胞的表面超微结构和功能的改变具有一定意义。
In this paper, polylysine cationic membrane preparation of human blood cells by scanning electron microscopy sample. Due to the interaction between the polyanionic surface of the blood cells and the polycation surface of the polylysine, various cells can be strongly adsorbed onto the glass plate pretreated with poly-L-lysine. When cells are in contact with the surface, cell attachment occurs immediately. Adsorbed cells stretched well, arranged in a single layer, evenly distributed, the picture is clear, easy to observe without losing the original morphological characteristics of the cell differential. The adsorbed biological material is not peeled off due to fixing, dehydration and critical point drying during preparation of the sample. Application of the method to prepare human peripheral blood lymphocytes, the Department of lymphocytes scanning electron microscopy observation samples. All of these cells showed a clear and strong three-dimensional scan image. This study of a variety of blood cells and other free cells on the surface ultrastructure and function of change is of significance.
