目的:建立同时测定清热消炎合剂中大黄素和大黄酚含量的方法。方法:采用高效液相色谱法,以VP-DOSC18(4.6mm×200mm,5μm)为色谱柱;以甲醇-0.1%磷酸溶液(80∶20)为流动相;检测波长为254nm;柱温为30℃;流速为0.8mL.min-1。结果:大黄素的回归方程为Y=4224748.8X+10068.6,r=0.9999(n=5),线性范围为2.5~25.0mg·L-1;大黄酚的回归方程为Y=5675274.7X+14297.8,r=0.9999(n=5),线性范围为3.0~30.0mg.L-1。结论:本方法可以同时测定样品中的大黄素和大黄酚的含量,方法简便,重复性好,回收率高,可用于该制剂质量控制。
Objective: To establish a method for simultaneous determination of emodin and chrysophanol in Qingre Xiaoyan Mixture. Methods: The mobile phase consisted of VP-DOSC18 (4.6 mm × 200 mm, 5 μm) column and methanol-0.1% phosphoric acid solution (80:20) as mobile phase at a detection wavelength of 254 nm. The column temperature was 30 ℃; flow rate of 0.8mL.min-1. Results: The regression equation of emodin was Y = 4224748.8X + 10068.6, r = 0.9999 (n = 5), the linear range was 2.5 ~ 25.0 mg · L-1. The regression equation of chrysophanol was Y = 5675274.7X + 14297.8, r = 0.9999 (n = 5) with a linear range of 3.0 to 30.0 mg.L-1. Conclusion: The method can simultaneously determine the content of emodin and chrysophanol in the sample. The method is simple and convenient, has good repeatability and high recovery, and can be used for quality control of the preparation.
