目的探讨微小RNA-218(miR-218)在上皮性卵巢癌细胞株SKOV3中发挥的生物学功能。方法采用实时定量PCR方法检测上皮性卵巢癌细胞株中miR-218的表达。通过转染miR-218模拟体、miR-218抑制剂外源性地增高或者降低miR-218在SKOV3中的表达,利用MTT、流式细胞术、癌细胞侵袭迁移实验观察miR-218对SKOV3增殖、凋亡和侵袭转移的作用。结果 miR-218能增强SKOV3增殖能力(P<0.05),而与细胞凋亡、侵袭转移无相关性(P>0.05)。结论 miR-218在上皮性卵巢癌细胞株SKOV3中发挥调节细胞增殖的作用。
Objective To investigate the biological function of microRNA-218 (miR-218) in epithelial ovarian cancer cell line SKOV3. Methods Real-time quantitative PCR was used to detect the expression of miR-218 in epithelial ovarian cancer cell lines. The expression of miR-218 in SKOV3 was exogenously increased or decreased by miR-218 inhibitor transfection with miR-218 mimetic. MTT, flow cytometry and invasion and migration of cancer cells were used to observe the proliferation of SKOV3 , The role of apoptosis and invasion and metastasis. Results miR-218 enhanced SKOV3 proliferation (P <0.05), but not apoptosis, invasion and metastasis (P> 0.05). Conclusion miR-218 plays an important role in the regulation of cell proliferation in epithelial ovarian cancer cell line SKOV3.
