探讨鼻咽癌 (nasopharyngeal carcinoma,NPC)组织中热应激蛋白 2 7(heat stress protein 2 7,HSP2 7) ,P5 3及 P2 1癌基因蛋白表达水平及其相互关系 ,采用间接免疫荧光染色和 SABC相结合的双重荧光染色法 ,分析 3种蛋白在鼻咽癌发生、发展及预后中的作用。结果发现 :32例 NPC中 HSP2 7,P5 3和 P2 1阳性表达比例 (阳性率 )分别为83.8%、 5 0 %、 93.8% ;8例鼻咽增生性炎症组织中三者比例为 4/ 8、 0 / 8、 4/ 8;4例正常组织则全部阴性表达。 HSP2 7、P5 3、 P2 1的共表达以 HSP2 7与 P2 1的共表达最为显著 ,且与 NPC恶性程度和临床分期有关。提示 :HSP2 7及 P2 1阳性表达可能是 NPC发生的早期变化 ,可将二者联合作为 NPC早期诊断及预防的敏感生物标志物。
To investigate the expression of heat stress protein 2 7 (HSP2 7), P5 3 and P2 1 oncogene proteins in nasopharyngeal carcinoma (NPC) tissues and their correlations. The expression of heat shock protein 2 7 SABC combination of double fluorescence staining, analysis of three kinds of protein in the occurrence, development and prognosis of nasopharyngeal carcinoma role. The results showed that the positive expression rates of HSP2 7, P5 3 and P2 1 in 32 NPC specimens were 83.8%, 50% and 93.8%, respectively. The proportions of the three in 8 nasopharyngeal proliferative inflammatory tissues were 4/8 , 0/8, 4/8; all four normal tissues showed negative expression. The co-expression of HSP2 7, P5 3 and P2 1 was the most significant in co-expression of HSP2 7 and P2 1, and was related to the malignancy and clinical stage of NPC. It is suggested that the positive expression of HSP27 and P2 1 may be the early changes of NPC, which can be used as sensitive biomarkers for the early diagnosis and prevention of NPC.
