目的:从细胞及分子水平分析低剂量米非司酮在子宫内膜对胚泡接受性中的作用.方法:以组织学方法观察内膜形态变化;免疫组织学方法测定LIF及IL-6在子宫内膜的基因表达以及用半定量反转录—聚合酶链反应(RT-PCR)测定内膜雌、孕激素受体nRNA水平等方法来评估子宫内膜的成熟度.结果:米非司酮在剂量为0.03mg·kg~(-1)·d~(-1)时,可明显延缓子宫内膜的成熟,抑制间质细胞蜕膜化,降底LIF和IL-6在内膜的基因表达和增高内膜雌、孕激素受体mRNA水平.结论:小剂量米非司酮可直接作用于子宫内膜,影响内膜的成熟而抑制内膜对胚泡的接受性.
OBJECTIVE: To analyze the effect of low dose mifepristone on the receptivity of endometrium to blastocyst at cellular and molecular level.Methods: The morphological changes of endometrium were observed by histology, and the expression of LIF and IL-6 Endometrial gene expression and semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) to determine endometrial estrogen and progesterone receptor nRNA levels and other methods to assess the endometrial maturity.Results: Ketone at a dose of 0.03 mg · kg -1 d -1 significantly delayed the maturation of endometrium, inhibited the decidualization of stromal cells, reduced the expression of LIF and IL-6 in the intima Gene expression and increase endometrial estrogen and progesterone receptor mRNA levels.Conclusion: A small dose of mifepristone can directly affect the endometrium, affecting endometrial maturation and inhibiting endometrial receptivity to blastocyst.
