试验以枯草芽胞杆菌ATCC6633株为材料,研究可诊断苯丙酮尿症(PKU)Guthrie检验用培养基的选择及拮抗剂(β-2-噻吩丙氮酸)的使用浓度及培养时间。试验结果表明,Demain培养基在所试3种培养基中最适于Guthrie检验;拮抗剂在Guthrie检验中的最佳使用浓度为1.50×10-5mol·L-1与3.00×10-5mol·L-1之间,并确定检验培养时间为16h。
In the present study, Bacillus subtilis ATCC6633 strain was used to study the selection of media for the detection of Guthrie in PKU and the concentration of antagonist (β-2-thienylazemic acid) and incubation time. The results showed that the Demain medium was the most suitable for the Guthrie test in the three kinds of medium tested. The optimum concentration of antagonist in the Guthrie test was 1.50 × 10-5mol·L-1 and 3.00 × 10-5mol · L-1, and to determine the test culture time of 16h.
