目的 探讨原发性肝细胞癌 (简称肝癌 )细胞凋亡相关蛋白 Bcl- 2和 Bax的表达及其意义 .方法 采用免疫组化ABC法检测 Bcl- 2和 Bax蛋白在 66例人肝癌组织和 2 4例人正常肝组织 ,以及 3株人肝癌细胞系 HCC- 92 0 4 ,SMMC-772 1 ,HHCC和 1株永生化的人正常肝细胞系 QZG的表达情况 ,并分析肝癌组织的 Bcl- 2和 Bax阳性率与其病理分级之间的关系 .结果 Bcl- 2在正常肝组织的阳性率只有 4.2 % ,而在肝癌组织为 2 5.8%差异显著 (P<0 .0 5) .Bax在正常肝组织的阳性率为 70 .8% ,在肝癌组织为 43.9% ,差异显著 (P<0 .0 5) .肝癌组织的 Bcl- 2和 Bax阳性率与其病理分级之间均未见明显的相关性 (P>0 .0 5) .在正常肝和肝癌组织中 ,Bcl- 2阳性率均明显低于 Bax(P<0 .0 1 ,P<0 .0 5) .在QZG细胞中未见明显的 Bcl- 2阳性信号 ,Bax阳性细胞计数率为 61 % .SMMC- 772 1细胞和 HHCC细胞的 Bcl- 2阳性细胞计数率分别为 1 4 .3%和 1 1 .8% ,Bax阳性细胞计数率分别为 58.3%和 47.1 % .HCC- 92 0 4细胞中未见明显的 Bcl- 2阳性信号 ,Bax阳性细胞计数率为 43.2 % .结论 与正常肝组织相比 ,肝癌组织的 Bcl- 2蛋白低度升高 ,Bax蛋白下降 .正常肝和肝癌组织的 Bcl- 2水平均低于 Bax.肝癌和正常肝细胞系的 Bcl- 2蛋白水平均很
Objective To investigate the expression of Bcl-2 and Bax in patients with primary hepatocellular carcinoma (HCC) and their significance.Methods Immunohistochemical ABC method was used to detect the expression of Bcl-2 and Bax protein in 66 cases of human hepatocellular carcinoma and 2 4 human normal liver tissues and 3 human hepatocellular carcinoma cell lines HCC-92 0 4, SMMC-772 1, HHCC and 1 immortalized human normal liver cell line QZG, and analyzed the expression of Bcl-2 And the positive rate of Bax and its pathological grade.Results The positive rate of Bcl-2 in normal liver tissue was only 4.2%, while it was 5.8% in liver cancer tissue (P <0.05) .The positive rate of Bax in normal liver The positive rate of HCC tissue was 70.8%, while it was 43.9% in HCC tissues (P <0.05). There was no significant correlation between the positive rate of Bcl-2 and Bax in HCC tissues and the pathological grade (P> 0.05) .The positive rates of Bcl-2 in normal liver and liver cancer tissues were significantly lower than those in Bax (P <0.01, P <0.05), but not in QZG cells Of Bcl-2 positive cells, the positive rate of Bax positive cells was 61% .The positive rate of Bcl-2 positive cells in SMMC-772 1 cells and HHCC cells were 14.3% and 11 .8%, the positive rate of Bax positive cells were 58.3% and 47.1% respectively.No obvious Bcl-2 positive signal was found in HCC-92 0 4 cells, the positive rate of Bax positive cells was 43.2% .Conclusion Compared with normal liver tissue , Bcl-2 protein in hepatocellular carcinoma increased and Bax protein decreased, Bcl-2 levels in normal liver and liver cancer tissues were lower than that in Bax.
