目的:观察苏拉明对脑膜瘤细胞增殖及周期的影响。方法:在无血清、10~(-8) MIGF-Ⅰ或含20%小牛血清条件下,递增浓度的苏拉明加入培养体系中,进行细胞计数和~3H-TdR摄取率测定,用流式细胞仪分析苏拉明对细胞周期的影响。结果:在不同情况下,10~(-4)M苏拉明显著抑制~3H-TdR掺合入DNA中(P<0.01),10~(-3)M苏拉明几乎完全阻断细胞的DNA合成;10~(-4)M苏拉明使细胞增殖下降47%~60%,并使S期和G_2/M期细胞比例显著增加(P<0.05和P<0.01)。结论:苏拉明在体外能有效抑制脑膜瘤细胞增殖,并使脑膜瘤细胞S期和G_2/M期延长或停滞。
Objective: To observe the effects of suramin on the proliferation and the cycle of meningioma cells. Methods: Suramin at increasing concentrations was added into the culture system under the condition of no serum, 10 ~ (-8) MIGF-Ⅰ or 20% fetal bovine serum, and the cell count and the uptake rate of ~ 3H-TdR were measured. Cytometric analysis of suramin on the cell cycle. Results: Under different conditions, suramin 10 ~ (-4) inhibited the incorporation of ~ 3H-TdR into DNA (P <0.01), and suramin 10 ~ (-3) M almost completely blocked cells DNA synthesis; 10 ~ (-4) M suramin decreased cell proliferation by 47% -60%, and significantly increased the proportion of cells in S phase and G 2 / M phase (P <0.05 and P <0.01). Conclusion: Suramin can effectively inhibit the proliferation of meningioma cells in vitro and prolong or arrest the S phase and G 2 / M phase in meningioma cells.
