目的:观察全身亚低温对大鼠全脑缺血再灌注损伤细胞凋亡的影响,探讨亚低温的脑复苏机制和脑缺血后凋亡刺激因素.方法:采用改良的Pulsineli-WA4VO法,制备大鼠全脑缺血再灌注模型,采用TUNEL法,观察海马脑区细胞凋亡的特征变化.结果:发现脑缺血20min再灌注6h,海马CA1区可见少量细胞核呈蓝染颗粒,于再灌注12h,24h,48h,3d,5d蓝染阳性细胞明显增多,5d达高峰,7d开始下降.给予亚低温治疗后上述变化明显减轻.结论:亚低温脑复苏作用可能是通过抑制或延迟脑缺血后细胞凋亡,而抑制或阻断凋亡发生的启动级联反应为其重要机制.
Objective: To observe the effects of systemic mild hypothermia on cell apoptosis after global cerebral ischemia-reperfusion in rats and to explore the mechanism of mild hypothermia in cerebral resuscitation and the factors of apoptosis after cerebral ischemia. Methods: A rat model of global cerebral ischemia-reperfusion was established by modified Pulsineli-WA4VO method. The changes of apoptosis in hippocampus were observed by TUNEL method. Results: After reperfusion for 20 min at 20 min of cerebral ischemia, a small amount of blue nuclei were observed in CA1 area of hippocampus. At the 12 h, 24 h, 48 h, 3 d and 5 d after reperfusion, the number of blue staining positive cells increased significantly, reaching the peak on the 5th day and decreasing on the 7th day. Mild hypothermia treatment to give the above changes significantly reduced. CONCLUSION: Mild hypothermic cerebral resuscitation may be an important mechanism by inhibiting or delaying cell apoptosis after cerebral ischemia and inhibiting or blocking the initiation cascade of apoptosis.
