目的研究炎性细胞因子刺激对人胚胎胰岛分泌功能的影响。方法采用常规消化法分离人胚胎胰岛并进行原代培养,分别加入rhTNFα、rhIL-1β、rhIFNγ或同时加入2种或3种细胞因子,培养24和48小时取其上清测定胰岛素、胰升糖素含量。结果3种细胞因子单独或联合应用均可不同程度地抑制胰岛素的分泌,联合应用对抑制胰岛素分泌有协同作用;对胰升精素的分泌则有明显的促进作用,但联合应用的协同作用因胰岛培养时间的不同而异。结论细胞团子影响胰岛细胞分泌功能的机制可能是复杂多样的。
Objective To investigate the effects of inflammatory cytokines on pancreatic islet secretion in human embryos. Methods The islets of human embryos were isolated by conventional digestion method and cultured in primary culture. Two or three cytokines were added to rhTNFα, rhIL-1β, rhIFNγ, respectively. The supernatants were cultured for 24 and 48 hours to measure insulin, Vegetarian content. Results The three cytokines alone or in combination could inhibit the secretion of insulin to a certain extent. The combined application of the three cytokines had a synergistic effect on the inhibition of insulin secretion and the promotion of secretion of pancreatic ascending hormone. However, the synergistic effect Islet culture time varies. Conclusion The mechanism by which the cell mass affects the secretory function of islet cells may be complex and diverse.
