研究酶解青蛤制备血管紧张素转化酶(Angiotensin-converting enzyme,ACE)抑制肽。以ACE抑制率为指标,先筛选最适酶种;探究温度、时间、p H、加酶量和料液比等因素对ACE活性的影响,通过L16(45)正交试验优化酶解工艺。结果表明,最适酶为胰蛋白酶,最佳制备工艺条件为:反应温度45℃,反应时间9 h,p H 8.0,加酶量1 200U/g,料液比为1︰2(g/m L)。最佳工艺所得酶解物的ACE抑制率为36.8%。为青蛤的高值化利用提供了理论依据。
To study the enzymatic hydrolysis of Cyclops clam to prepare angiotensin-converting enzyme (ACE) inhibitory peptide. The ACE inhibition rate was used as an index to select the most suitable enzyme. The effects of temperature, time, pH, amount of enzyme and solid-liquid ratio on ACE activity were investigated. The enzymolysis process was optimized by L16 (45) orthogonal test. The results showed that the optimal enzyme was trypsin. The optimal conditions for the preparation were as follows: reaction temperature 45 ℃, reaction time 9 h, pH 8.0, enzyme dosage 1200 U / g, solid - liquid ratio 1︰2 (g / m L). The ACE inhibitory rate of the best enzymatic hydrolyzate was 36.8%. It provides a theoretical basis for the high value utilization of Cyclops.
