目的:研究醒脑静注射液对酒精性昏迷大鼠外侧下丘脑区(LHA)谷氨酸和γ-氨基丁酸的影响。方法:腹腔注射乙醇制作酒精性昏迷大鼠模型,20min后侧脑室给药,微透析技术收集透析样品(每30 min收集一次),OPA柱前衍生化法处理样品,高效液相色谱(Synergi Hydro-RP C18,150 mm×4.6 mm,4μm),荧光检测脑脊液中谷氨酸(Glu)、γ-氨基丁酸(GABA)的含量。结果:与模型对照组比较,醒脑静注射液0.34 mg/kg、0.68 mg/kg、1.37 mg/kg外侧下丘脑区(LHA)Glu、GABA恢复到正常水平的时间更短。结论:醒脑静注射液能够通过调节酒精性昏迷大鼠外侧下丘脑区(LHA)Glu、GABA的含量,使其恢复到正常水平从而促进觉醒。
Objective: To investigate the effects of xingnaojing injection on glutamate and γ-aminobutyric acid in the lateral hypothalamus (LHA) of alcohol-induced coma in rats. Methods: Alcoholic coma was induced in rats by intraperitoneal injection of ethanol. Intracerebroventricular administration was given 20 minutes later. Dialysis samples (collected every 30 minutes) were collected by microdialysis technique. OPA pre-column derivatization was used to treat the samples. Synergi Hydro -RP C18, 150 mm × 4.6 mm, 4 μm). The levels of glutamic acid (Glu) and γ-aminobutyric acid (GABA) in cerebrospinal fluid were detected by fluorescence. RESULTS: Compared with the model control group, the time of returning to normal level of Xingnaojing Injection 0.34 mg / kg, 0.68 mg / kg and 1.37 mg / kg LHA Glu and GABA was shorter. Conclusion: Xingnaojing injection can promote the awakening by regulating the level of Glu and GABA in the hypothalamus (LHA) of rats with alcohol induced coma and returning them to normal levels.
