为研究青鱼生长激素基因工程菌在豆粕中的发酵工艺,试验采用L25(56)正交设计探讨温度、pH、培养时间、豆粕质量浓度、接种量和诱导剂质量浓度等6个因素对青鱼生长激素基因工程菌的生长和蛋白表达量的影响。结果表明:各因子对蛋白表达量的影响明显,它们的作用大小依次为发酵时间>pH>接种量>发酵温度>诱导剂质量浓度>豆粕质量浓度,最优发酵组合条件为发酵时间96h、pH7、接种量5%、发酵温度30℃、诱导剂质量浓度1%及豆粕质量浓度6%。确立发酵工艺后进行10L发酵罐发酵,活菌数达到79.6亿CFU/mL,青鱼生长激素表达量占总蛋白的28.4%。
