目的:探讨降脂合剂对高脂血症大鼠血脂谱及血液流变学的影响,阐明降脂合剂调节血脂及血流动力学的作用机制。方法:将雄性Wistar大鼠60只随机分为正常对照组、模型对照组、辛伐他汀组和降脂合剂高、中、低剂量组,每组10只。除正常对照组用基础饲料喂养外,其余各组大鼠喂饲高脂饲料建立高脂血症大鼠模型,造模同时预防给药,空白对照组及模型对照组大鼠给予生理盐水2mL灌胃,辛伐他汀组大鼠给予7.2×10-4g.mL-1的辛伐他汀混悬溶液2mL灌胃,降脂合剂低、中、高剂量组大鼠分别给予0.25、0.50、1.00g.mL-1的中药煎剂2mL灌胃,以上给药均为每日1次,持续喂养及给药10周。10周后6组大鼠均摘眼球取血,全自动生化分析仪测定大鼠血脂谱,全自动血液流变分析仪测定大鼠血液流变学的变化。结果:与模型组比较,降脂合剂各剂量组大鼠预防用药后,胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)和载脂蛋白B(ApoB)的水平均不同程度降低,高密度脂蛋白(HDL-C)、载脂蛋白A(ApoA)的水平及apoA/apoB比值均有不同程度的升高,差异均有统计学意义(P<0.05或P<0.01);与模型组比较,降脂合剂各组大鼠血浆黏度(ηp)、全血黏度(ηb)(低、中、高切)、红细胞聚集指数(IEA)、红细胞压积(HCT)及红细胞电泳时间(EET)降低,红细胞变形指数(IED)升高,差异均有统计学意义(P<0.05或P<0.01)。结论:降脂合剂能够调节实验性高脂血症大鼠血脂代谢及血液的黏、聚、凝状态,对血脂和血流异常有较好的调节和抑制作用。
Objective: To investigate the effects of Jiangzhi Mixture on blood lipid profile and hemorheology in hyperlipidemic rats and to elucidate the mechanism of Jiangzhi Mixture in regulating blood lipid and hemodynamics. Methods: Sixty male Wistar rats were randomly divided into normal control group, model control group, simvastatin group and hypolipidemic mixture high, medium and low dose group, 10 rats in each group. In addition to the normal control group fed with basal diet, the other groups of rats fed high-fat diet to establish hyperlipidemic rat model, while modeling prevention and control, blank control group and model control group rats given saline 2mL irrigation The rats in the stomach and simvastatin group were administered with 2 mL simvastatin suspension solution of 7.2 × 10-4 g · mL-1 orally. After 10 weeks, the rats in all the 6 groups were taken out of their eyes for blood collection. The blood lipid profile was measured by automatic biochemical analyzer and the hemorheology of rats was measured by automatic hemorheology analyzer. Results: Compared with the model group, the levels of cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL-C) and apolipoprotein B The level of HDL-C, ApoA and the ratio of apoA / apoB were all increased in varying degrees, the differences were statistically significant (P <0.05 or P < 0.01). Compared with the model group, the plasma viscosity (ηp), whole blood viscosity (ηb) (low, middle and high), erythrocyte aggregation index (IEA), hematocrit (HCT) The time of erythrocyte electrophoresis (EET) decreased and the index of erythrocyte deformability (IED) increased. The difference was statistically significant (P <0.05 or P <0.01). CONCLUSION: Jiangzhi Mixture can regulate blood lipid metabolism and blood viscosity, aggregation and coagulation status in experimental hyperlipidemic rats and has good regulation and inhibition on blood lipid and blood flow abnormalities.
