目的通过探讨参附注射液中间体及不同配比对新生大鼠心肌细胞膜ATP酶活性及细胞内相关离子的影响,揭示参附注射液两种中间体最佳配比。方法通过胰蛋白酶消化法和差速贴壁法获得心肌细胞,分别给予附子提取物30μl·ml-1、红参提取物30μl·ml-1及附子:红参1∶2、2∶1、1∶1不同配比药液,作用1h,检测心肌细胞的细胞活力、ATP酶的活性及相关离子的浓度。结果参附注射液中间体及各配比药液对心肌细胞的细胞活力,Ca2+-ATP酶、Ca2+-Mg2+-ATP酶及Na+-K+-ATP酶的活性及细胞内Na+、Ca2+、K+、Mg2+的浓度产生不同影响,其中以附子:红参2:1作用最强,且与前期研究中参附注射液的作用一致,均能提高心肌细胞的细胞活力,提高Ca2+-ATP酶、Ca2+-Mg2+-ATP酶及Na+-K+-ATP酶的活性,降低细胞内Na+、Ca2+的浓度,提高Mg2+、K+的浓度。结论参附注射液中间体及不同配比均对正常心肌细胞具有保护作用,作用以附子:红参2∶1最强。
Objective To explore the influence of Shenfu injection on the membrane ATPase activity and intracellular related ions in neonatal rats by exploring the intermediates and different proportions of Shenfu injection and to reveal the optimal ratio of the two intermediates. Methods Cardiomyocytes were obtained by trypsin digestion and differential adherent method. The aconitine extract 30μl · ml-1, red ginseng extract 30μl · ml-1 and aconite: red ginseng 1: 2, 2:1, 1 : 1 different proportion of liquid, the role of 1h, detection of myocardial cell viability, ATPase activity and related ion concentrations. Results The activity of Ca2 + -ATPase, Ca2 + -Mg2 + -ATPase and Na + -K + -ATPase and the activity of intracellular Na +, Ca2 +, K +, Mg2 + Concentration of aconite: red ginseng 2: 1 has the strongest effect, and with the effect of Shenfu injection in the previous study, all can improve the cell viability of cardiac myocytes and increase the activity of Ca2 + -ATPase, Ca2 + -Mg2 + -ATPase and Na + -K + -ATPase activity, reduce the concentration of Na + and Ca2 + in cells, and increase the concentration of Mg2 + and K +. Conclusion Shenfu injection intermediates and different proportions of normal cardiomyocytes have a protective effect, the role of aconite: red ginseng 2: 1 strongest.
